SPOLA Technology

VARIOM’s proprietary Single-Phase Oligonucleotide Ligation Assay (SPOLA) is a method for the detection of mutations in nucleic acids. The technology is of a diagnostic grade but can also be applied in SNP validation studies in research laboratories.

Using our proprietary production method, target oligonucleotides are attached to 96-well microplates. This format confers flexibility in assay content and the potential of high throughput genotyping using standard automation technology.

powerful
- up to 48 genotypes per multiplex-assay in a standard 96-well microplate format
- almost any sequence context accessible

reliable and accurate
- greater than 99% accuracy due to the dependence on two hybridisation reactions and a ligation step

rapid

- completed in little more than 1 hour post-PCR
simple
- common microplate format
- uniform test procedure for all assays
- performed on the lab bench at room temperature
- minimal hands-on time and equipment requirements
- quantitation with standard plate readers or simple visual scoring with clear yes/no results

scalable
- manual, semi-automated or fully automatable with standard plate handling instrumentation for throughputs of thousands of genotypes per day
- up to 48 separate bi-allelic loci can be tested per plate.

cost-efficient
- no capital expenditure
- no training costs and simple interpretation
- competitive price
- no third party costs

accessible
- preconfigured assays available for most relevant mutations in single and multiplex panel formats

flexible
- fast turnaround times for customized assays
- microplate format allows content flexibility
