SPOLA Technology

Using our proprietary photochemical production method, detection oligonucleotides are attached to polystyrene carrier materials, provide the surface on which the Solid-Phase Oligonucleotide Ligation Assay (SPOLA) takes place. Applied to planar arrays, bead arrays or VARIOM’s current platform, the SPOLA is;

Reliable and accurate
- Greater than 99% accuracy due to the dependence on two hybridisation reactions and a ligation step

Flexible
- Almost any sequence context accessible
- Can detect not only SNPs but small insertions, deletions and substitutions

Simple

- Unique reaction conditions allow assay to take place at room temperature as opposed the thermal cycling step in standard Oligonucleotide Ligation Assay.

In addition, our current platform, the 96-well microplate, confers distinct advantages;

Powerful
- Up to 48 genotypes per multiplex-assay in a standard 96-well microplate format

Scalable
- Manual, semi-automated or fully automatable for throughputs of up to thousands of genotypes per day

Flexibility in multiplexing
- Up to 48 separate bi-allelic loci can be tested per plate

Simple
- Industry standard microplate format
- Uniform test procedure for all assays
- Performed on the lab bench at room temperature
- Minimal hands-on time
- Instrument-free protocol
- Results quantified with standard plate readers or visual scoring with clear yes/no results

Economical
- No capital expenditure
- No training costs and simple interpretation
- Competitive price
- No third party costs
Rapid
- Completed in little more than 1 hour post-PCR
Accessible
- Preconfigured assays available for most relevant mutations in single or multiplex panel formats
